What is PNC-28 Peptide?

PNC-28 is a synthetic peptide derived from the mdm-2-binding domain of the p53 protein, specifically residues 17-26. It includes a penetratin sequence that facilitates cellular penetration, making it effective in inducing necrosis in various cancer cell lines without affecting normal cells.

PNC-28 Research as an Anti-Cancer Peptide

Research has shown that PNC-28 is effective in inducing necrosis, rather than apoptosis, in various human cancer cell lines, including those lacking the p53 gene, which is often mutated in cancers. This action is attributed to the peptide’s ability to bind to the hdm-2 protein on cancer cell membranes, leading to the formation of pores that disrupt the cells and cause necrosis [R, R, R, R].
